Lab D.4 Week 1 November 6, 2023 1. What are Gemini Viruses (GV) and what makes them unique and easier to study than most other viruses? Which GV are we employing in this lab? Gemini Viruses are a classification of plant viruses that have single-stranded DNA unlike most plant viruses which have RNA genomes. Gemini viruses are easier to study than most other viruses because their DNA genome is more stable than RNA genome. In this lab, we will find out if the studied disease produces a new genome and if the new genome is a Gemini Virus 2. Why might one use an agarose gel as opposed to an acrylamide gel in this experiment? An agarose gel has a large pore size which helps separate duplex DNA strands and large proteins/molecules. 3. Provide an outline or flowchart for this two-week experiment. i. Purification of plant DNA ii. Gel electrophoresis of plant DNA iii. Neutralize gel? iv. Southern Blot transfer v.
