Cassia auriculata L. (CA) is profoundly used as a tonic, astringent, anti-diabetic, conjunctivitis and opthalmia in Ayurvedic medicine (Subhadradevi et al., 2011). It is one of the principle constituent of Avaarai panchaga chooranam; an Indian herbal formulation used in the treatment of diabetes to control the blood sugar level (Berman et al., 2001). Cassia species, are rich sources of polyphenols, anthraquinone derviatives, flavanoids, polysaccrides, saponins, tannins and steroids (Singh et al., 1980; Yen et al., 1998; Ayo et al., 2004), flavanoids, sterols/terpenoids and phenolic acids are known to be antidiabetic bioactive principles. Flavanoids are known to regenerate the damaged β cells in alloxan induced diabetic rats…show more content… Adult albino rats of wistar strain (180-240 g) of either sex were housed at 12h light/dark cycle at room temperature (27±2°C) and fed for 24 h with food and water ad libitum. The approved protocol (JSSCP/PhD/PH CHEM/06/2012-13) was conducted in accordance with guidelinesas per “Guide for the care and use of Laboratory animals”.
2.5. Induction and assessment of diabetes for Pharmacodynamic study
The overnight fasted adult albino rats of wistar strain weighing 180-240 g were induced diabetes by single intra-peritoneal injection of streptozotocin (45mg/kg), prepared by dissolving in citrate buffer (0.1 M pH 4.4). The fasting glucose levels was witnessed using One touch glucometer (Accu-Chek Active). The inception value of fasting glucose level to diagnose diabetes was considered to be more than 250mg/dL after 48 hours and used for pharmacodynamic study. The animals were gulfed into three groups entailing six animals each. Group 1 animal received the MT higher dose 90mg/kg p.o (MT-HD); Group 2 received MT-HD along with CA 500mg/kg p.o (MET-HD-CA); Group 3 received MT Lower dose 45mg/kgalong with CA 500mg/kg p.o (MET-LD-CA). After administration, blood samples were collected on daily basis for 21 days from retrio orbital sinus under ether anesthesia and glucose levels were estimated.
2.6 Pharmacokinetic study design
The non-diabetic, adult albino rats of wistar strain weighing 180-240g, the overnight fasted animals were gulfed into three groups with six animals
