There are two main types of Plasmodiumthat infect humans

Plasmodium Falciparum

Plasmodium Vivax

Transmitted by female mosquitoes

Develops in mosquito gut

Migrates to salivary glands

Transfers to other organisms through the saliva of the mosquito.

A mosquito is an organism of the family Culicidae. The females require a blood meal to develop eggs.

In this test, I will be using Plasmodium Berghi, a type of malaria that is only infectious to mice.


Anopheles stephensi mosquitoes, both eggs and live mosquitoes


Anti-Pbs21 gene in plasmids


Lots and lots of eppendorf tubes


Airtight habitat for mosquitoes

Random assorted materials


Inserting plasmid into mosquito egg

Bacterial transformation

Inserting plasmid into Asaia


Infecting mosquitoes with transformed Asaia

Transgenic mosquito

Take plasmids containing anti-Pbs21 gene and insert them into mosquito eggs using a microinjector. Repeat several times to increase the chances of getting a successful mosquito.

Grow the mosquito to maturity.

Asaia-infected mosquito

Take Asaiabacteria and put it with plasmids containing the anti-Pbs21 gene and the gene for GFP

Heat shock the bacteria to make them accept the plasmids.

Plate the bacteria and select the colonies that glow in the UV light

Infect adult mosquitoes with the successful bacteria.

Take both types of mosquito, as well as normal mosquitoes as a control, and set them up in exactly equal habitats.

Evaluate each generation of mosquito on

Percentage of ones with immunity to malaria
