• The Primase is the key in DNA replication because no known DNA polymerases can initiate the synthesis of a DNA strand without the initial DNA. It also acts like a halting mechanism to prevent the leading strand from outpacing the lagging strand by halting the progression of the replication fork.
• Helicase attach to and break apart the hydrogen bonds between the bases on the DNA strands hereby pulling apart the two strands. As a result providing a single-stranded DNA for replication, transcription, and recombination.
• Ligase is essential for putting together okazaki fragments during replication, and also for completing short- patched DNA synthesis occurring in DNA repair process.
