Any chemical or physical agent which changes the structure of DNA is termed as mutant. 5-Bromouracil is a type of mutagen which cause mutation in the nucleotide sequence of DNA. Chemically it is a bromine containing compound which is structurally like thymine.

5-BrU exists in three tautomeric forms which pairs with different nucleotides. The three forms are keto, enol and ion forms. The base-pairing properties of these forms depends on their chemical structure. 5-BrU is an analog of thymine thus it normally pairs with adenine. The amount of 5-BrU in enol form is higher as compared to thymine because of the presence of more electronegative Br atom. The Br atom is more electronegative than methyl group thus, it can easily cause alterations in base sequence of DNA.
