Step 1

In microbiology, the stock solutions i.e. those solutions containing  high concentration of microorganism is usually diluted so as to get the colony forming units (cfu) in countable range. 

If stock is plated directly, the cfu would be so large that it cannot be counted accurately. So, in such cases, the stock is serially diluted and dilutions are plated. The cfu count of these dilutions is observed and hence, the dilution in which cfu's can be easily and accurately counted is used to quantify the cfu of stock. 

In case of virus, such as bacteriophage, colonies are not formed rather plaques does. So, the result of the plate is reported as plaque forming units or pfu.

Step 2

Given, pfu of 10-6 dilution of bacteriophage  = 46 pfu

This count is by using 0.1 mL of 10-6 dilution. So, pfu/mL at this dilution = 46/0.1 = 460 pfu/mL
