How is phenyl isothiocyanate useful in determining protein structure?

Expert Answer

In determining protein structure, The first step is to determine the amino acid composition of the peptide.

The peptide is heated in 6 N HCl at 110°C for 24 hours. This helps in obtaining the amino acids that the peptide is made of.

The next step is to identify the N-terminal amino acid.

Reaction of Phenyl isothiocyanate:

Phenyl isothiocyanate reacts with the uncharged termina...
