HYDROLYSIS OF DNA
A. To the fibrous mass, add 10 ml of 10% H,SO, in a beaker. Boil gently or 5 minutes. Perform
the following test:
1. Inorganic phosphate – to 1 ml NH,OH, add 1 ml filtrate. Acidify by placing a small piece
of blue litmus paper using 10% HNO, until the litmus paper changes to red. Then add 2 ml
ammonium molybdate. Boil the solution and allow to stand for a few minutes. Note color of
ppt.
2. Purines - to 2 ml filtrate add 3 ml of 3M NH,OH. Add 2-3 drops of 0.1M AgNO,
solution to it. Note color of precipitate.
3. Deoxyribose – to 1 ml filtrate add 1 ml of diphenylamine reagent. Heat for 10 mins in a
boiling water bath. Cool and note color produced.
