Tumor suppressor genes regulate a cell cycle during division and replication. If the cell grows uncontrollably the TSG will check the defect and stop the cells from division. If the TSG has loss of function mutation in both the alleles then it will allow the cell to grow abnormally. This can cause development of cancer. They are grouped into caretaker and caregiver genes.
