Proteins are the organic compound composed of 20 or more amino acids joined by peptide bonds.

There are many tests like electron microscopy, NMR spectroscopy, and X-ray crystallography to determine if a protein is a membrane protein or not.

The feature of protein structure is the balance between hydrophobic and hydrophilic and the structure stability depends heavily on the molecule's hydrophobic effects. The determination of the hydrophobic value of amino acid is mainly calculated according to the distribution coefficient in which various amino acids are in organic solvent and water. Due to the different laboratory test equipment, experimental conditions, organic solvents, and calculation methods, hydrophobicity values obtained are also of a considerable gap. When we map these amino acids sequence of protein onto a sequence of hydrophobicity, we need to optimize a variety of different hydrophobic parameters.

Both prokaryotic and eukaryotic cells are used to determine the membrane protein among the other proteins.
