Chapter 12, Problem 1SAQ
Summary Introduction

To explain: The type of chromophore associated with a negative stain.

Introduction: Bacteria are unicellular prokaryotic organisms that maintain a definite shape since they contain the rigid cell wall. They are classified into different types based on the structure, namely coccus (spherical shaped), Bacillus (rod-shaped), filamentous, spirilla (spiral-shaped), appendaged, and pleomorphic bacteria. The structure of bacteria can be viewed under the microscope using the staining methods.

Explanation of Solution

The stain is the organic compound that consists of an auxochrome group and chromophore group. The chromophore is the color imparting compound that binds to the charged cellular components such as proteins, nucleic acids, and so on. The negative stain is the acidic stain that imparts a negative charge on the chromophore after dissociation and hence, it binds to the positive charge of the bacterial cell. Hence, it is anionic in nature.
