To explain: The reasons why agar is stabbed several times with the loop when the bacteria from the throat swab are streaked on the agar plate.
Introduction: The initial identification of the streptococcal and enterococcal species is based on their hemolytic pattern on their growth on the blood agar. Certain species of these two groups destroy the red blood cells on the blood agar whereas certain species would partially break down the hemoglobin and certain species do not exhibit hemolysis on the blood agar.
Explanation of Solution
Blood agar is used for cultivation of fastidious organisms such as streptococcal species. The blood agar is used for determining the hemolytic capability of the microorganisms. The blood agar should be stabbed several times with the loop while streaking the bacterial sample obtained from the throat swab. This is because stabbing on the blood agar would facilitate the anaerobic environment for the streptococcal species to grow. Moreover, stabbing would also improve the progression of hemolysis.
