Fred Sanger discovered the dideoxy method, where adenine, guanine, cytosine, and thymine in varying length are produced. The four DNA fragments (adenine, guanine, cytosine, and thymine) are radio labeled and separated through electrophoresis. For the Sanger sequencing, the separated fragments needs different reactions and different electrophoresis gel for each one of them, where the sequence can be determined.

A copy of single stranded DNA to form a sequence is determined by the use of DNA polymerase enzymes. The four fragmented DNA is grown with the aid of DNA polymerase that utilizing the deoxyribonucleoside triphosphate as a substrate and followed adding of different dideoxy analogs. The elongation of the DNA fragments is terminated by the dideoxy analog, as they lack 3’-hydroxyl. Hence, the different dideoxy analog acts a chain terminator for the nucleotide bases.
