Standard Preparation:
1.
Weigh 10 mg of the Acetaminophen RS accurately and dissolve in 15 ml of methanol. Shake well.
2.
Add 85ml of water to adjust the volume to 100 ml.
3.
Pipet 5 ml from the stock solution and dilute to 50 ml with water.
Test Preparation:
1.
Weigh the 10 tablets individually and record on the worksheet provided.
Powder 10 tablets and weigh a portion of the powders equivalent to 100mg.
2.
3.
Transfer the powders into 100 ml VF.
4.
Add 15 ml of methanol, shake to dissolve and dilute with water to the volume to make 100 ml.
5.
Pipet 1 ml from the previous solution, transfer it into a 100-ml volumetric flask and dilute with water to volume.
Instrument:
UV-Vis Spectrophotometer
Blank: water
Cell- 1cm quarts cell
Wavelength:243 nm
Procedure:
1.
Turn on the power switch. Allow the instruments to warm up for at least 20 minutes to stabilize the source and detector
2.
After the warm up period, set the desired wavelength using the UP and DOWN arrows.
3.
Using the MODE Key, Select the operating mode as TRANSMITTANCE, ABSORBANCE or CONCENTRATION.
Choose appropriate cuvettes both for standard and sample solutions.
4.
5.
Fill up a clean cuvette with a blank solution and wipe dry the outer surface to remove droplets, dust or fingerprints.
Place the cuvette in the Blank (first) compartment and align the guide mark on the cell. Place the cell firmly and close
6.
the lid. Use the other positions for sample solutions to be measured.
7.
Adjust the meter to 100% T or 0 Absorbance reading by pressing 0 Abs/100%T key until display reads "100.0%T" or
0.000A.
8.
Pull or push the cuvette selector arm and put the cuvette containing the sample solution in front of the light beam and
read the %T or Abs value of the samples.
Record the Absorbance readings.
9.
10. Repeat the procedure until absorption of all solutions is observed.
11. When all measurements are completed, empty and clean the cuvette and turn off the device.
