Explain the principles of next-generation sequencing and metagenomics

Expert Solution
Principle of next-generation sequencing

Next-generation sequencing

The principle of Next Generation Sequencing is same to Sanger sequencing, which very much depends on capillary electrophoresis. The genomic strand is first fragmented, and then bases in each fragment are identified by emitted signals when these fragments are ligated against a template strand. The Next generation sequencing method use array-based sequencing. Array based sequencing combines the techniques which were developed in Sanger sequencing to process millions of reactions in parallel, resulting in very high speed and throughput at a reduced cost
