uncontrolled cell proliferation, as in cancers, is primarily due to deregulation of the cell cycle.  

CDKs are cell cycle regulators that phosphorylate to activate consequent retinoblastoma protein so as to promote progression in the cell cycle. however, it also needs tight controlling which is brought about by Cdc25 phosphatase which dephosphorylates CDKs which lifts the inhibition of these molecules.
