Question

You followed all the steps correctly with a kit you bought last week. However, NO purified DNA came out of the miniprep (More than one answer possible).

a) The plasmid was lost during culture growth
b) Your tears from doing the same experiment again and again contaminated the reagents, causing them not to work
c) Depurination of the DNA led to loss of genetic material
d) Incomplete lysis of the cells after adding lysis buffer
Expert Solution
Step 1 Introduction

Plasmids exist naturally in bacterial cells . In some cases bacteria need to pick up plasmid from the environment , as they readily lose them during the antibiotic resistance and during the process of conjugation , as one of the daughter cell may not receive plasmid .

Depurination involves the loss of purine bases from DNA . In this process N- glycosyl bound to deoxyribose is broken by hydrolysis , leaving the DNA sugar -phosphate chain intact , producing an absic site.
