A single molecule of DNA will have four types of nucleotide bases, adenine (A), guanine (G), cytosine (C), and thymine (T). Here, adenine and guanine are purine while thymine and cytosine are pyrimidine bases. Using this Chargaff’s has put forward a rule called base pairing rule for double stranded DNA. The adenine will always pair with thymine and guanine with cytosine. So the composition of purines (A+G) will always be equal to that or pyrimidines (T+C). This is the first base pair rule.
