The enzyme Lysozyme is present in human nasal mucus and catalyzes the breakdown of a component of the cell wall of certain bacteria, causing them to burst. Hence, Lysozyme helps protect against bacterial infections. One experimental investigation of Lysozyme gave the result in Table 1. Here, [S]o is the initial substrate concentration and V is the initial rate of the breakdown reaction. [S]O (µM) 1.0 2.0 4.0 10 20 Table 1 V (μm/sec) 0.14 0.22 0.31 0.42 0.48 Use a graphical method to determine the Michaelis constant, KM, and the maximal rate, Vmax. What initial reaction rate would be expected for [S]o=200μM? In these experiments the concentration of lysozyme was 0.0025 µM. Calculate the maximal turnover, kcat, for lysozyme under the investigated conditions. Lysozyme belongs to Enzyme Class number 3 (EC 3), the so-called hydrolases. What characterizes a hydrolase?

Question

THIS IS THE SECOND TIME IM POSTING THIS QUESTION! I'm trying to figure out how to calculate WITHOUT using computer (its not allowed during exam). I need to do what they ask me to do without computer. So please, dont answear this question with a plot from Excel or just some MM equations without any grapgh showing the work.

I will appreciate an answear for what they ask in the exercise, and nothing else. Please, details when explaining this exercise, so I can understand it. Thank you

Follow-up Question

There is actually something im confused about, when you calculating V0 for S0=200uM --> V = 0.48*200/ 2.42 + 1

Why are you only using the S0 value=200uM one place in the equation and then using the S0 value from given data the second place in the equation?

Shouldnt S0 be 200 uM both places in the equation?

Follow-up Question

You are absolutely the BEST. Finally someone that dosen't calculate this exercise as 836f66dgsffjgvkik!!!!

I understand everything you did, but I have one question.

Some experts says that km can be calculated by saying Vmax from date divided by 2 and then they find Km by going down on x-axis on the graph. But, you used the MM equation, to solve for Km. Is both method the same? Or is one of them more precise than the other?

*And if theres no need to reply back after you answering my question, then I will thank you again for your help. I have more these types of exam questions that I will probably upload later, and I hope someone like you will look at it.
