1 Lab 3: Porifera and Cnidaria Objectives 1. Compare similarities and differences in the structures of sponges and cnidarians. 2. Investigate evolutionary differences for achieving the same survival goals. 3. Introduce how to use the dichotomous key to identify various taxa of Porifera and Cnidaria. Learning Goals To identify differences in the structure of sponges and cnidarians. To investigate evolutionary differences for achieving the same survival goals. To use a dichotomous key to identify sponges and cnidarians. To learn how to pick the best microscope for viewing a specimen. Introduction Today we are going to investigate two phyla within the Kingdom Animalia: Phylum Porifera (sponges) and Phylum Cnidaria (jellyfish and their relatives). Remember all animals are multicellular eukaryotes without cell walls. They are also all heterotrophs who ingest their food rather than absorb it. Animals are predators, herbivores, and detritivores. In addition all animals are motile at some point in their life cycle. They are capable of moving from one location to another under their own power although some lose this capability as adults. Only animals have muscle and nerve tissues although these tissues are not present in all animals. Safety Handle glass microscope slides with care. Use gloves while handling preserved specimens. Never put dissected specimens in the trash can, put them in the designated containers.
2 Experiment Using the materials provided: 1. Each person must use their own microscope for the microscope slides. 2. Clean the lens and the slides using lens paper. 3. Work with a pairs to observe the preserved specimens. 4. In your lab notebook draw one of each specimen provided. You must do your drawings from the preserved and microscope specimens not from illustrations. The drawings should be large even when the specimens are small. 5. Usually you should draw the specimen using the high dry objective unless it is too large to see all of it properly, in that case, use the low dry objective. 6. Using the dichotomous key on the last page, write out the steps to key the sponge, sea anemone, and H ydra’s phylum in you lab notebook. Available Tools and Equipment 1. Microscope: light and dissecting 2. Lens Paper 3. Dissection tray with forceps 4. Preserved sponge ( Grantia ) 5. Grantia c.s. (sponge) microscope slide 6. Preserved Sea Anemone 7. Metridium c.s. & l.s. (sea anemone) microscope slide 8. Hydra w.m. with bud microscope slide 9. Hydra w.m. with ovary microscope slide 10. Hydra w.m. with spermaries microscope slide Important Notes to Include With Your Drawings 1. Using a dissection microscope, observe a preserved sponge and label the osculum 2. Pick a slide labeled Grantia c.s. (sponge) microscope slide, draw the entire cross section and label the spongocoel and choanocytes.
