Hypothesis:
If we streak on different media plates (R2A, LB, 10% TSA and MacC), we could
possibly see different levels of growth as well as different colors, as MacConkey is selective and
differential.
Protocol:
1.
Label petri dish on the bottom, not the lid.
2.
Obtain sterile toothpick and collect a small bacteria sample from the plate.
3.
Streak the four different media plates (R2A, LB, 10% TSA, and MacC) with the isolated colonies
that had somewhat a zone of inhibition using the aseptic laboratory technique.
4.
Store plates upside down and refrigerate them until the next lab.

We see effective growth on all forms of plates. LB, TSA, R2A all showed strong growth. On the
MacC plate, we see a reddish pink plate color, indicating a lactose fermenting colony. The
growth on MacC is minimal, but the little growth we see is bright red in color.
Conclusion: We conclude that by streaking the isolated colonies with zones of inhibition, we
saw different levels of growth depending on the plate. From most growth to least growth, the list
includes LB, 10% TSA, R2A, and then MacC. Since we saw growth on the MacC plate, we are
able to say that the colony we streaked is gram negative. We also saw color change to a
reddish/pink color, which indicates lactose fermentation.
