LAB #8
Presumptive Testing of Stains
Objective:
Which presumptive test is need for the stain identifies characteristics of the body fluid and is it is positive
presumptions for the Prescence of the type of body fluid depending on their test.
Complete Test steps Procedures:
Kastle-Meyer (KM)Test
:
detects presence of
Blood
Procedure:
-
Moisten the swab with distilled water
-
Swab the stain with the moisten cotton swab
-
1-2 drops ethanol from a pipet
-
1-2 drops of phenolphthalein from a pipet
-
1-2 drops of 3% hydrogen peroxide
Acid Phosphatase (AP) Test
:
detects presence of
Seminal Material
Procedure:
-
Moisten the filter paper with distilled water
-
Lay on to the entire stain, and press it down with the palm of your hand for 15 seconds
-
Turn the paper over and pour the AP reagent
-
13 seconds and it turns purple
Chloride Test
: detects the presence of
Urine
Procedure:
-
3 drops of distilled water are added to the stain
-
1 drop of 2% nitric acid
-
1 drop of silver nitrate
-
Cloudy reaction
Ultra-Violet Light fluorescence
:
Below describe the type of fluorescence for each fluid:
Blood:
No
Saliva:
Yes
Seminal Material:
Yes
Urine:
Yes
Water:
Yes

:
Description of specimen, noting all observed Stain(s) by color and location.
Next,
Diagram the specimen; note each stain indicating each with a stain number.
Next,
For each & every Stain……..next to the stain #, list the following:
1. Characteristics of stain,
2.
Test performed with the test result
What the test result means;
What the next required serological test is.
Spec #1 is
Small brown paper bag contains white paper towel from the kitchen(sc). It’s folded, with two reddish
brown stains detected. Mirror image stain when folded.
