Procedure A: Culture Medium Preparation
You will prepare LB (Luria Broth) media, which is a standard complex media that is used to

1. Determine mass of powder(s) to be used. Every litre (IL) of LB media contains the
following:
10 g Bacto-tryptone
• 5 g yeast extract
10 g NaCl
**Each group of two students should make 150 mL of this media. Calculate the masses of
reagents you will need.
2. Weigh out the reagent(s). Weigh the required amount of powder on a balance and add it to
an empty beaker.
3. Add water. Add water to the beaker to a level just BELOW the final volume of your
solution. (For example, if you want to make 150 mL of solution, you should add water to
just BELOW the 150 mL mark on the beaker.)
4. Stir. Add a stir bar and stir until the powder complete dissolves in the water.
