Pre-Lab Lab 1: Microbiology Lab Safety (100 points ( · Each question on the lab worksheet must be answered completely, thoroughly, in complete sen- tences, and correctly in order to be considered for full credit · If the question asks you to do research or find a source, a reputable, credible, and/or scholarly source citation must be included in order to be considered for full credit · If a math formula is required to arrive at an answer, work must be shown otherwise, no credit will be awarded Questions 1. What constitutes personal protective equipment? When should personal protective equipment be worn? Why is personal protective equipment important? (5 points) Personal protective equipment includes Safety Goggles/ Glasses, Gloves, and Lab coat/Apron . They should be worn the whole time while doing lab experiments and in the lab . Personal protective equipment is very important to protect people from safety hazards . 2. As part of the lab safety procedure and before beginning an experiment, what must you do in order to be prepared to conduct the steps of the experiment? (5 points) Read each procedure completely before starting. You should be family with the actions required every step of the way. Prepare a workplace by clearing the area of all clutter, food, and other items not directly related to the laboratory exercise. Read the labels on all chemicals and note their stay rating, if you have any questions to concerns about chemicals, refer to the DS available at the eScience Lab website. Consult your physician if you are pregnant allergic to chemicals, or have other medical conditions that may require additional protective measures. Remove all loose clothing and always closed-toe shoes; pull back and secure long hair, remove all jewelry; wear safety glasses at all times, wearing soft contact lenses while conducting laboratory exercises is discouraged as they can absorb potentially harmful chemicals. When handling chemicals, always wear the protective glasses, gloves, and apron provided in your safety kit. 3. Why are there increasing levels of biological containment procedures? What level of biological containment will be necessary to complete the experiments in this manual? (10 points) a.The biosafety level describes the minimum required containment measures that should be used to ensure safety when working with microorganisms. The BSL is determined by the characteristics of the microorganism being studied and includes the level of knowledge of the Updated 9-2023 DM Microbiology Lab Safety
microorganism, association with disease, mode of transmission, and the existence of the disease . b. Only BSL-1 procedures will be used in these experiments . 4. What document should you refer to if you have questions regarding the safety or disposal of a chemical? Where can this document be located? (10 points). Refer to the DS available at the eScience Lab website . 5. Explain what the presence of dark to gray-colored fuzzy colonies indicates on growth plates. (10 points) Fuzzy-appearing growth indicates mold contamination Experiment 1 Data Tables Table 2: Experiment 1 Colony Growth (10 points ( Plate Condition Growth Day 1 Growth Day 2 Growth Day 3 Growth Day 4 Growth Day 5 1 Hand without washing 0 1 11 111 1111 2 Hand with washing 0 0 1 11 111 3 Yeast 0 0 1 1 1 4 Deionized water 0 0 0 1 1 Image 1 (10 points ( · Image: Insert an image of your growth plate #1 (when it has colonies present). · Identifier: Include your identifier clearly visible and written on a piece of white paper, your name, student ID, and the current, submitted date included in the image with the growth plate. ) No credit for this lab will be provided unless both the growth plate and identifier are included in the same image and can be clearly identified .( Updated 9-2023 DM Microbiology Lab Safety
