Isoelectric point is the pH at which the net charge is zero. pI is calculated as the average of two nearby pka values. Proteins are polymer of amino acids linked by peptide bond/amide bond (covalent) with release of a water molecule. Alpha carbon of amino acids contains -carboxyl group, amine group, side chain group and hydrogen atom. Based on the side chain group amino acids can be classified as polar, uncharged polar and non-polar.
