Eosin Methylene Blue (EMB) agar medium is generally used for the examination of E. coli. This medium acts as both selective and differential medium that is used to differentiate between coliforms and fecal coliforms. Since E. coli is a lactose fermenter, it ferments the lactose present in the EMB agar medium and produces metallic green color in the medium, and other non-lactose fermenters appear as colorless. Thus, the lack of strong reaction on the EMB medium plate refutes the determination that the isolated bacteria are an E. coli culture. However, other biochemical tests can be used to confirm the presence or absence of E.coli in the medium.
