ELN – Analytical Technique Extraction Extraction Purpose Demonstrate the use of extraction as a method of separating mixtures of compounds Notes - The stopcock is closed when it perpendicular to the flask Protocol Sample Preparation Weigh 2.0g of the substance into 125 Erlenmeyer flask. Record the exact weight Dissolve the substance into 40mL of solvent Assembly of the Apparatus Set up ring clamp, used to hold the separatory funnel, and place beaker underneath Pour the solution onto the funnel Slowly add 15mL of 6M hydrochloric acid to the separatory funnel. Extraction Insert the stopper in the separatory funnel and invert Cradle the funnel in the palm and hold the stopper between your fingers Point funnel upwards and towards the back of the hood and vent slowly by opening and closing the stopcock a few times. Gently shake the funnel and mix the immiscible liquids Allow layers to separate Remove the stopper Separate the liquids by opening the stopcock and draining the lower aqueous layer into a 125mL Erlenmeyer flask. Close the stopcock until the organic layer drains into another Erlenmeyer flask Put EtOAc back into the sep. funnel and repeat extraction with 15mL of fresh 6M HCl Drain the two phases as described previously and combine this aqueous phase with the first Erlenmeyer flask Recover Product Neutralize the solution Check the pH Allow the solution to cool and put in the ice bath before collecting using vacuum filtration Waste Throw away weighed paper in the appropriate waste container Rinse glassware with acetone afterward Aqueous phases are to be disposed of in the sink and EtOAc sol. in the appropriate container. 1
