Chapter 21, Problem 1CCS

Two patients—a woman and her husband, ages 23 and 22, respectively—arrive at the health clinic one morning. They report having had severe abdominal cramps, grossly bloody diarrhea, nausea, and fever for 48 hours. Cultures of stool samples grown under microaerophilic, capneic conditions contain comma-shaped, Gram-negative bacilli. Both the patients are lactovegetarians and report being part of a “cow leasing” program at a local dairy in which patrons lease part of a cow’s milk production so that they can drink natural, whole, raw milk. The couple devised the program so that they and several neighbors could circumvent state regulations prohibiting the sale of unpasteurized milk. Investigators obtained and cultured a milk sample from the dairy’s bulk milk tank: the cultures contained the bacterium pictured.

  1. 1. What is the pathogen?
  2. 2. How did the couple become infected?
  3. 3. Are the couple's colleagues at work at risk of acquiring an infection from the couple?
  4. 4. What other foods that are common sources of this bacterium can be ruled out in this case?

Below is a scenario requiring the use of laboratory equipment and instruments.    A watery fecal sample is tested for the presence of Bacillus cereus and Vibrio cholerae. The sample is stored in a screw capped tube. You are tasked to grow the Bacillus in a solid medium while Vibrio in a liquid medium.   Choices: A. Incubator, B Hanging Drop Slide , C.Petri Dish, D. Drying Oven, E. Bunsen Burner, F. Inoculating NeedlE , G. Volumetric Pipette, H. Screw-capped Tubes, I. Hot plate, J. Centrifuge, K. Inoculating Loop, L. Microscope, M. Beaker, N. Biosafety Cabinet   1. What could be used to determine if there are more bacteria in the sample aside from Bacillus and Vibrio?   2. What should you use if you want to observe the natural motility of Bacillus under a Brightfield microscope?   3. Where should you perform the task of sterilization and inoculation? 4.What should be used to transfer the sample to the broth?   5. What could be used to separate the solid from the liquid portion of the…
Based on the pictures provided, tell me what I have for the gram-positive, and what I have for the gram-negative? Possible Unknown Strains: Staphylococcus epidermidis, Bacillus subtilis, Staphylococcus aureus, Bacillus megaterium, Eschericia coli, Pseudomonas aeruginosa Name one negative and one positive of which I have.
A stool specimen of a patient with severe diarrhea was cultured in a series of specialized media for isolation of enteric organisms. The cultures yielded three isolates that were species of Salmonella, Shigella, and Escherichia. Explain why the Hydrogen sulfide production test would be diagnostically significant.
