Chapter 9, Problem 2VI
Summary Introduction
To answer:
The effectiveness of the brass pitchers with microcidal action.
Introduction:
Heavy metals such as copper, silver, zinc, and mercury are considered to have antimicrobial effects. There is a high density seen in the ions of the heavy metals. These metals have the ability to attract the microbes and bond with them. Once the microbes are bonded, the protein denaturation occurs, thus microbes are eradicated.

A microbiologist sets up a serial dilution to analyze bacteria in raw milk.
Tube 1 – 0.1 mL milk diluted in 9.9 mL sterile water
Tube 2 – 0.1 mL milk from Tube 1 diluted in 9.9 mL sterile water
Tube 3 – 1.0 mL milk from Tube 2 diluted in 9.0 mL sterile water
What is the dilution of raw milk in Tube 3?
In 1928, Sir Alexander Fleming was studying Staphylococcus bacteria growing in culture dishes. He noticed that a mold called Penicillium was also growing in some of the dishes. A clear area existed around the mold because all the bacteria that had grown in this area had died. In the culture dishes without the mold, no clear areas were present.
Fleming hypothesized that the mold must be producing a chemical that killed the bacteria. He decided to isolate this substance and test it to see if it would kill bacteria. Fleming transferred the mold to a nutrient broth solution. This solution contained all the materials the mold needed to grow. After the mold grew, he removed it from the nutrient broth. Fleming then added the nutrient broth in which the mold had grown to a culture of bacteria. He observed that the bacteria died which was later used to develop antibiotics used to treat a variety of diseases. (Biology Corner)

You obtain a 1 mL sample of contaminated drinking water in order to calculate the number of fecal bacteria in the water. The 1 mL sample is diluted into a first bottle with 99 mL of sterile saline. One mL from the first bottle is transferred into a second bottle of saline containing 9 mL. Finally, 1 mL from the second bottle is transferred to a third bottle with 9 mL of saline. You create two spread plates from the third bottle: plating 0.1 mL results in 106 colonies, while plating 1 mL results in 562 colonies. How many fecal bacteria per mL are in the drinking water?

- Bacterial growth depends on many environmental factors, including the temperature of the environment. Since microbes can survive in a certain range of temperatures and will thrive at a temperature, understanding these variables allows for control of their growth. This understanding can be used, for example, to preserve certain foods or to treat infections. This lab simulation will use an instrument called a spectrophotometer. This instrument quantitatively measures the amount of light that is absorbed or transmitted by molecules in solution. Microbial populations grow at different rates based on temperature. One way to measure growth is by turbidity measurements.
