To analyze:
Question asked for the description for comparative level Transcriptional and translational fusion of reporter gene with regulatory gene. The question also asked to discuss the difference in expression patterns of the two transgenes.
Introduction:
In eukaryotic, mRNA is modified in the nucleus, and they are exported from the nucleus to the cytoplasm for translation. During protein production, there are two important regulatory steps - transcription and translation. The cell has a mechanism which directs the translation of mRNA. Transcription of gene takes place but every transcribed gene does not get translated.

- Flowering Locus C (FLC) is a gene that is responsible for flowering in certain plants. FLC is normally expressed in new seedlings, which prevents flowering. When a plant is exposed to cold temperatures, FLC expression decreases and the plant produces flowers. FLC is regulated through epigenetic modifications. Based on this information, you should expect which of the following in histone proteins near the FLC gene: 1.In new seedlings, histones will be acetylated; while in the cold, histone methylation occurs 2.In new seedlings, histones will be methylated; while in the cold, histone acetylation occurs 3.In new seedlings, histones will be methylated; while in the cold, histone deacetylation occursarrow_forwardAssume that there is a double stranded break on DNA double helix of an eukaryotic cell due to X-ray radiation and it is not repaired. In addition, the cell’s Apaf-1 protein is not expressed due to a null mutation in the Apaf-1 gene. Please discuss the effect of not having Apaf-1 expression in the cell with non-repaired double stranded break.arrow_forwardHelp me pleasearrow_forward
- GR and PPAR are transcription factors that bind to GRE and PPARE sequences respectively and activate transcription of genes. A reporter cell line is created in which the the green fluoresecent protein (GFP) is controlled by a GRE sequence and the pink fluorescent protein mCherry is under control of a PPARE sequence. If the gene for GR is introduced into the reporter cell line, the cells produce a green color. Chimeric proteins are created in which the DNA Binding Domains (DBD) and Activation Domains (AD) of the transcription factors are introduced into various cell lines. Match the following cell-types with the fluorescent color(s) you would expect the cells to produce.arrow_forwardWhat role does an operator sequence serve in bacterial gene expression regulation? Describe one change in bacterial cells that can cause a repressor protein to go from inactive to active or from active to inactive.arrow_forwardHistone methylation can have many different effects on gene expression. In some cases, histone methylation is associated with activation of transcription, whereas in other cases it can trigger the formation of heterochromatin and a decrease in transcription. If histone methylation has been detected in the region of gene YFG in yeast, describe an experiment that could distinguish whether the methylation is important to activate or repress transcription of gene YFG.arrow_forward
- Termicin is a small antifungal protein in termites that is produced by cells and secreted into termite saliva in response to a pathogen. In vitro translation of the termicin-encoding gene is performed, and the effects of that product are compared to those of termicin extracted from a termite. You see that extracted termicin exhibits more antifungal behavior than in vitro translated termicin. After further analysis, you see that extracted termicin contains 3 disulfide bonds, while in vitro translated termicin contains zero. The addition of microsomes to the in vitro translation reaction results in termicin with all 3 disulfide bonds. What experimental condition is most likely responsible for this difference? A. in vitro translation was not performed at the correct temperature affecting protein folding B. a mutation occurred during in vitro translation, leading to differences in disulfide bond formation O C. the UPR can not be activated in vitro, therefore, this protein can only be…arrow_forwardDesign a transcriptional circuit that regulates the expression of β-galactosidase in a eukaryoticcell, such that the enzyme is produced in the presence of lactose and repressed in the absenceof lactose. Use at least one of the following elements: histone acetyltransferase, histonedeacetylase, chromatin remodeling complexarrow_forwardIn a qPCR experiment, you find that the mRNA for the gene CDK1 goes down (decreased expression) when you treat your cells with doxorubicin. You wonder whether this is due to epigenetic changes on the gene for CDK1. Name two methods that you might use to check whether there are changes in epigenetic modifications (e.g. DNA methylation or histone tail modifications) on the CDK1 DNA.arrow_forward
- Transcriptional repressor proteins (e.g., lac repressor), antisense RNA, and feedback inhibition are three different mechanisms that turn off the expression of genes and gene products. Which of these three mechanisms will be most effective in each of the following situations? A. Shutting down the synthesis of a polypeptide B. Shutting down the synthesis of mRNA C. Shutting off the function of a protein For your answers to parts A–C that list more than one mechanism, which mechanism will be the fastest or the most efficient?arrow_forwardWhat are the functions of transcriptional activator proteins and repressor proteins? Explain how they work at the molecular level.arrow_forwardDiscuss the following argument: “if the expression of every gene depends on a set of transcription regulators, then the expression of these regulators must also depend on the expression of other regulators, and their expression must depend on the expression of still other regulators, and so on. cells would therefore need an infinite number of genes, most of which would code for transcription regulators.” how does the cell get by without having to achieve the impossible?arrow_forward
