Topic: Enzymes
Catalase Lab
Background Information:
Virtually all of the biochemical reactions that occur in living organisms are regulated by enzymes. Enzymes are proteins that function as biological catalysts, acting on a particular substrate to increase the rate of a particular type of reaction within an optimal set of conditions. One source of enzymes is the liver, which breaks down many substances within the body. Catalase, one of the enzymes found in the liver, breaks down hydrogen peroxide (H2O2), a toxic waste product of cellular metabolism. It must be disposed of because it is highly reactive and can damage DNA and interfere with the proper functioning of many cellular enzymes. Catalase speeds up the decomposition of hydrogen peroxide into water and oxygen gas. The enzyme, catalase, can also be found in hard fruits or vegetables such as (apples, potato or turnips), which you will be exploring in this lab.

I. Active site analysis.
Below is a diagram of a putative active site for Monoamine oxidase. As we learned, the purpose of
tertiary structure is to form a scaffold so you can orient just a few amino acids in the right orientation
to promote binding and/or catalysis. The position where this occurs is the active site. The amino acid
architecture of an active site is designed to bind substrates. Amino acid side chains are capable of
hydrogen bonding, ionic and hydrophobic interactions. Fill in each amino acid that you think is
suitable for interacting with the part of the substrate it is closest to. Assume the pH will be at 7.0
