Concept introduction: When two molecules of an ester undergo condensation is called claisen condensation. Like in aldol reaction, in claisen condensation one molecule is a nucleophile and the other molecule is an electrophile.
A claisen condensation reaction is a reversible reaction and occurs in the presence of a strong base.
The first step of claisen condensation is the removal of a proton from alpha carbon by a base and then the addition of enolate ion formed to the second ester molecule forming a β-ketoester.
A crossed-claisen condensation is also a condensation reaction between a ketone and an ester. Primarily one product will form from crossed-claisen condensation when one of the ketone or easter do not have any alpha hydrogens.

- The widely used anticoagulant warfarin (see Chemical Connections: From Moldy Clover to a Blood Thinner in Chapter 18) is synthesized from 4-hydroxycoumarin, benzaldehyde, and acetone as shown in this retrosynthesis. Show how warfarin is synthesized from these reagents.arrow_forwardWhen a primary alcohol is treated with p-toluenesulfonyl chloride at room temperature in the presence of an organic base such as pyridine, a tosylate is formed. When the same reaction is carried out at higher temperature, an alkyl chloride is often formed. Explain.arrow_forwardHow many different Claisen condensation products are possible in the reaction of equal amounts of ethyl acetate (CH3CO2Et) and ethyl propanoate (CH3CH2CO2Et)?arrow_forward
- 2-Ethyl-1-hexanol was needed for the synthesis of the sunscreen octyl p-methylcinnamate. Show how this alcohol could be synthesized (a) by an aldol condensation of butanal and (b) by a malonic ester synthesis starting with diethyl malonate.arrow_forwardAcetals are often prepared during the synthetic sequence to protect the carbonyl group of an aldehyde or ketone. Such an acetal is not reactive to each of the following reagents except: a.) LiAlH4 b.) PCC(Collins) c.) H3O+ d.) RMgBr e.) NaOHarrow_forwardThe compound eutypine is an antibacterial agent isolated from the fungus Eutypa lata. This fungus results in a disease common to vineyards called eutyposis. Give a sequence of reactions that will take the following reactant and give eutypine when the other reactants used in the sequence are acetylene and acetone.arrow_forward
- Claisen condensation between diethyl phthalate and ethyl acetate followed by saponification, acidification, and decarboxylation forms a diketone, C9H6O2. Propose structural formulas for compounds A and B and the diketone.arrow_forwardThe compound 3,5,5-trimethyl-2-cyclohexenone can be synthesized using acetone and ethyl acetoacetate as sources of carbon atoms. New carbon-carbon bonds in this synthesis are formed by a combination of aldol reactions and Michael reactions. Show reagents and conditions by which this synthesis might be accomplished.arrow_forward
