Concept Introduction:

Restriction endonucleases recognizes a specific nucleotide sequence in a DNA double strand and cut the double stranded DNA at a particular position. This leads to the formation of two segments of double stranded DNA if the original DNA was linear, otherwise, if it was a circular DNA, cleavage leads to a linear double stranded DNA. All these DNA fragments have two ends with known sequence. These ends are known as sticky ends. As the name implies that there are unpaired nucleotides which are capable of making hydrogen bond readily with another segment of DNA which has complementary bases.
