Deoxyadenosine methylase is commonly referred to as DAM methylase. This is an enzyme that catalyzes the addition of methyl ( CH3 ) group to the adenine of the sequence of 5'GATC3' in the DNA (Deoxyribonucleic acid) which is newly synthesized.

The process of addition of methyl group to the adenine of the sequence of 5'GATC3' in the DNA can be referred to as DNA methylation. The process of DNA methylation stabilizes the newly synthesized DNA as well as help in the development of embryo and cell differentiation.

During the process of replication, methylated DNA strand remains temporarily unmethylated. Thus, in the two strands of DNA, one strand of DNA remains methylated, while the other strand of DNA remains unmethylated. This situation is referred to as hem-methylation. The unmethylated strand of DNA quickly methylates in the presence of enzyme dam methylase.

In the deficiency of enzyme dam methylase, DNA is left completely unmethylated. Also, the unmethylated DNA has more probability to undergo mutation. Thus, the deficiency of dam methylase enzyme increases the spontaneous mutation rates.
