BIOL 2071 Introductory Microbiology & Techniques – Bonus lab assignment Name: Malek Bleibel Sec. No.: 53 Results from Lab 10 – Bacteriophage cultivation and bioluminescent bacteria Max 10 marks Bacteriophage titre 1. Fill in the table below using your data (from posted images in Brightspace). [3 marks] Plaque assay results: Dilution Number of plaques Control (no phages added) 0 10 -2 TMTC 10 -3 TMTC 10 -4 TMTC 10 -5 TMTC 10 -6 32 10 -7 TLTC 2. Calculate the PFU/ml of the original phage solution: [2 marks] PFU/ml = (#of plaques * dilution factor)/ amount plated PFU/ml= (32*10 6 )/0.05 =6.4x10 8 PFU/ml 3. If you have a plaque assay plate that appears to have no plaques, what might this indicate? What else should you look for with such a plate? [2 marks] If no plaques are shown on a plaque assay this probably means that there was not enough phage concentration that infected the bacteria. However, it could also mean that there was too much phage that there were no unaffected bacteria left to define the plaques. In a plate with no plaques you should look for bacterial overgrowth which could suppress plaque formation. Bioluminescent bacteria 4. How did your “lux art” turn out? Was there anything that surprised you, that you might do differently if you were to do this again, or advice you’d give to another student? [1 mark] 1
BIOL 2071 Introductory Microbiology & Techniques – Bonus lab assignment My lux art turned out pretty good, I was a little disappointed that my results were upside down which is something I recommend you pay attention to when it’s being carried out, but it was pretty cool and the color is amazing. General lab reflection 5. What is a concept/fact that you learned in this lab, or a skill/technique you’ve developed that will be useful to you in future? Explain how/where it might be relevant or helpful. [2 marks] One concept that is I learned in this lab that can be helpful in the future is carrying out aseptic technique. Even though that might look different in different labs the idea of making sure to carry out everything in a manner that prevents any contamination is always useful. END OF ASSIGNMENT 2
