Chapter 19, Problem 10TYU
Summary Introduction
To explain: How neuraminidase inhibitor drug Oseltamivir (Tamiflu) helps in prevention of flu or reduces disease course of flu in a flu patient.
Introduction: An antiviral drug, Oseltamivir (Tamiflu), is recommended for patients infected by the influenza virus. Influenza virus is an RNA virus belonging to the family of Orthomyxoviridae. This virus produces flu with symptoms such as fever, sore throat, and coughing. Oseltamivir is a drug that inhibits the enzyme neuraminidase.

You think you need may have found a new inhibitor (NI) of the HIV protease, so you performed few experiments to characterize its behavior. You found that if you add an excess of substrate to the protease in the presence of the inhibitor, enzyme activity decreased at low substrate but reached the same vmax as observed in the absence of the inhibitor. Also, the affinity of the substrate for the enzyme is decreased by the inhibitor. What class of inhibitor is NI, competitive, uncompetitive, non competitive, or mixed type?
In the Avery experiment, mice were killed if they injected with S strain cells. Furthermore, incubation with extract from dead S strain cells could transform non pathogenic cells into pathogenic cells. This effect of S strain extract was eliminated if S strain extract is treated with what enzyme before incubation with non-pathogenic cells?
protease
DNAse
RNAse
Lipase
AZT (zidovudine) inhibits the synthesis of the HIV virus RNA because AZT resembles substrate molecules. Which kind of inhibition is most likely taking place in this reaction?

The company is starting a high throughput screen of potential drug targets against ecoDam I, a DNA adenine methyltransferase. Even more exciting, it might also be able to inhibit antibiotic resistant bacteria.(1) After the first round of screening, you and your supervisors have identified several potential targets. Your group went back to the lab to do more detailed enzyme kinetic inhibition analysis. You are going to need to analyze your enzyme kinetic data to see which of these inhibitors is best. Specifically, you need to see which of them inhibits at the lowest concentration, and determine their inhibition…arrow_forward
