Before sequencing the primary structure of an amino acid, it is first subjected to heating in 6 M HCl at 100–110oC for at least 12–36 hours for hydrolyzing the peptide bonds. Then these hydrolysates are separated from the mixture with the help of either HPLC (high-performance liquid chromatography) or by the ion exchange chromatography column.
