Chapter 8, Problem 5MTF
Summary Introduction

Introduction

The gel electrophoresis method is used to separate the genetic material or PCR amplified products based on their molecular size, shape, and electrical charge. This technique helps to identify and isolate genes of interest or DNA fragments. Such DNA sequences are used to insert into donor organisms through vectors to create genetically modified organisms (GMOs) or serve as a template for PCR amplification or construction of the gene library. The electrophoresis technique is involved to evaluate PCR samples for clinical or genetic diagnosis. Also, it is used for the separation of restricted DNA or RNA samples before transfer in Southern blotting or Northern blotting, respectively.

Dilution Problem.A culture of Staphylococcus is diluted as follows:(1) 70mL are added to 330mL of water.(2) 100uL from (1) are then added to 9.9mL of water.(3) A 10-3 dilution is made from tube # (2).(4) 10uL from (3) are plated for a pour plate and incubated. There were 80 colonies counted on one half of the plate following incubation. a) What was the overall dilution?b) How many cfu/mL were present in the original culture?c) How many milliliters of water is needed to make a 10-1 dilution using 100 uL from the original culture?
serology (elisa) lab: If the sample gave a negative result for the disease-causing agent, does this mean that you do not have the disease? What reasons could there be for a negative result when you actually do have the disease?
