Chapter 9, Problem 3TMW
Summary Introduction
To determine
The reason behind the use of Bacillus endospores as sterility indicators.
Introduction:
Endospores of bacteria such as Bacillus are used as biological indicator of sterility. The endospores of bacteria are impregnated into tape and after autoclaving; the tape is aseptically inoculated into sterile broth. The original material is said to be sterile, if there is no bacterial growth appears.

What makes heat an effective sterilizing agent?
Why are the conditions required for sterilization greater for dry heat (160-180°C for 1-2hrs) than for steam (121°C for 15min)?
Why does motility media contain a lower amount of agar than ordinary agar?
What concentration of agar is used in a semisolid medium for motility determination? How does that compare to a typical solid medium ? Explain.
