Chapter 12, Problem 15PDQ

Examples of histone modifications are acetylation (by histone acetyltransferase, or HAT), which is often linked to gene activation, and deacetylation (by histone deacetylases, or HDACs), which often leads to gene silencing typical of heterochromatin. Such heterochromatinization is initiated from a nucleation site and spreads bidirectionally until encountering boundaries that delimit the silenced areas. Recall from earlier in the text (see Chapter 4) the brief discussion of position effect, where repositioning of the w+ allele in Drosophila by translocation or inversion near heterochromatin produces intermittent w+ activity. In the heterozygous state (w+/w), a variegated eye is produced, with white and red patches. How might one explain position-effect variegation in terms of histone acetylation and/or deacetylation?

Typically, increased acetylation and decreased methylation of histone N-terminal tails in nucleosomes leads to removal of the protein components of the chromatin from the DNA relaxed packaging of the chromatin and increased transcription increased amounts of euchromatin relative to heterochromatin activation of topoisomerase tighter packaging of the chromatin and reduced transcription
Describe the packaging of chromosomal DNA by histones with diagrammatic representations. Name the various histone modifications and describe any two among them.
In the first figure of “DNMT3L Connects Unmethylated Lysine 4 of Histone H3 to de novo Methylation of DNA,” the authors determined that the DNMT3L protein interacts with several histone proteins. Using Figure a and b (from that paper), explain how each of the 4 histones interact with DNMT3L.
