Question
Chapter 14, Problem 9P
Interpretation Introduction
Interpretation:
The reason for Fura-2 to be ineffective for the study of calcium levels in living cells needs to be explained.
Concept introduction:
Fura-2 is a ratiometric and sensitive indicator dye. Thisis used for measuring calcium within the living cells. One of the forms of Fura-2 is known as acetoxymethyl (AM) ester. it is very helpful for noninvasive intracellular loadings. This is also available inpackages (F-1221) and in a solution with DMSO (F-1225).

Polyisocyanates are involved in the induction of the full spectrum of hypersensitivity responses and nonimmune inflammatory reactions including asthma. Which of the following statement is not correct with respect to these observations and impact on immunity?
a. A person once sensitized to polyisocyanate can demonstrate group reactivity to this set of compounds.
b. Individuals having an acute exposure to toluene diisocyanate is more likely to develop hypersensitivity then individuals chronically exposed to low vapor levels.
c. A mechanism for this hypersensitivity is the toluene diisocyanate forms a covalent bond with proteins that are part of the airway resulting in the immune system developing a response to “self” proteins.
d. Adaptive immunity can interact or “message” the Innate immune system to up or down regulate its expression.
e. All of the statements are correct.
Inducers and Inhibitors of AEP.
Short peptides such as legumain stabilization and activity modulation (LSAM) domain and αvβ3 integrin could enhance the activity of AEP. LSAM domain known as the prodomain of AEP blocks substrate binding before activation. This prodomain has a helical structure and two independent peptides. One is an activation peptide (AP, K287 to N323), and the other is a LSAM domain. LSAM domain remains even after AP is cleaved and released from protease at neutral pH via electrostatic interaction. AEP without LSAM domain has a lower melting temperature than AEP with LSAM domain [77, 117]. Another short peptide, αvβ3 integrin, can directly interact with AEP, and after forming a complex, the optimal pH for AEP activity is increased from 5.5 to 6.0. It indicates that αvβ3 binding could induce conformational stabilization of AEP accompanied by deprotonated C189. αvβ3 does not directly interact with the AEP active site; however, AEP docks to the αvβ3 RGD-binding site…
