Introduction: Melting temperature (Tm) is the temperature at which half of the DNA melts or denatures due to the breakage in the hydrogen bond that holds the two strands together. The melting temperature of a DNA duplex makes changes in the structure and stability of the DNA. The melting temperature of a DNA double helix depends on the number and percentage of base pairs present in the structure. An increase in temperature leads to hyperchromic effect on the polynucleotide.
