To explain: The difference between a gain-of-function mutation and a dominant-negative mutation.
Concept introduction:
Mutations are classified by their impact on the function of protein. The complete loss of function leads to the reduction of protein’s ability to work. The gain of function increases the protein function, and interferes with the wild-type protein’s function. The gain-of function mutation leads to the alternation of gene product processes a new pattern of gene expression. Dominant negative mutation refer to the defective gene interferes with the wild-type copy functioning.
