Chapter 10.2, Problem 5TQ
Summary Introduction

To review:

The expression of araBAD in the absence of AraC.

Introduction:

AraC (arabinose C) is a regulatory protein that activates or represses transcription and expression of an operon. The operon is called araBAD, as it includes three structural genes of arabinose operon. The AraC functions by activating or repressing the genes encoding for arabinose. In the presence of arabinose, it is broken down into an intermediate product called D-xylulose-5-phosphate.

What would happen if the operator sequence of the lac operon contained a mutation that prevented the repressor protein from binding the operator?  (Explain what would happen both in the presence and absence of lactose)
If glucose is not available, but lactose is available from the environment, what is the status of transcription of the lac operon genes? Explain your answer from both an evolutionary perspective and in terms of negative and positive regulation of the operon?
If glucose levels in the cell are high and lactose is available from the environment, what is the status of transcription of the lac operon genes? Explain your answer from both an evolutionary perspective and in terms of negative and positive regulation of the operon?
