Chapter 12, Problem 37P

The electrophoresis gel shown in part (a) is from a DNase footprint analysis of an operon transcription control region. DNA sequence analysis of a 35-bp region is shown in part (b). The control region, labeled with 32 P at one end, is shown in a map in part (c). Separate samples of control-region DNA are exposed to DNase I, and the resulting DNase I-digested DNA is run in separate lanes of the electrophoresis gel. Unprotected DNA is in lane 1 , DNA protected by repressor protien is in lane 2 , and RNA polymerase - protected DNA is in lane 3 . The numbers along the elctrophoresis gel corrospond to the 35-bp sequence labeled on the map in part (c). Use the information provided to solve the following problems.

Chapter 12, Problem 37P, 12.37 The electrophoresis gel shown in part (a) is from a DNase footprint analysis of an operon

a. Determine the DNA sequence of the region examined.

b. Locate the regions of the sequence protected by repressor protein and by RNA polymerase.

Parts A-C show the genotypes for partial diploid strains of the lac operon. Gene names have been simplified (ie, LacZ is shown as Z), with alleles shown as standard notations. There are two copies of the operon, one line is the copy in the genome and the other line is on a plasmid. In the blanks, indicate the following based on regulation of the structural gene in the header, choose "functional' if you expect that any amount of normal enzyme will be made, "nonfunctional" if you expect that transcription occurs but the enzyme is mutated, or "no transcription if the structural gene is not transcribed. Assume glucose is absent. LacY Enzyme Activity Genotype of Strain Absence of allolactose I-P+O+Z+Y+ I+P+ O° Z-Y- 1+ P+ OF Z+Y- PP+O+Z-Y+ 1-P+O+Z+Y- I+P-O+Z-Y+ Presence of allolactose
