Chapter 24, Problem 29P
Interpretation Introduction

Interpretation:

Alkylation of cysteine258 substituted aspartate aminotransferase gives some enzymatic activity.

Concept introduction:

Aspartate aminotransferase (AST) enzyme is found in all the cells of the body, mostly in heart and liver cells. This enzyme is very important in amino acid metabolism because it catalyzes the interconversion of aspartate and glutamate. During the conversion of a-ketoglutarate to glutamate, glutamate undergoes deamination t form ammonium. Reversely, aspartate is formed from OAA, which is used in the Krebs cycle.

Chemical labeling of chymotrypsin by the compound tosylphenylalanine chloromethyl ketone (TPCK) modifies the His 57 in the enzyme's active site. The structure of this derivative is shown below. TPCK inactivates the enzyme because the bulky addition prevents it from cleaving nearby covalent bonds. HCI + CH, C-O Chymotrypsin-His 57 TPCK Modified enzyme True O False
