The clinical symptoms of two forms of galactosemia—deficiency of galactokinase or of UDPglucose: galactose 1-phosphate uridylyltransferase—show radically different severity. Although both types produce gastric discomfort after milk ingestion, deficiency of the transferase also leads to liver, kidney, spleen, and brain dysfunction and eventual death. What products accumulate in the blood and tissues with each type of enzyme deficiency? Estimate the relative toxicities of these products from the above information.
Phosphoglycerate mutase (PGM) catalyzes the interconversion of 3-phosphoglycerate (3PG) and 2-phosphoglycerate (2PG) in the glycolytic and gluconeogenic pathways.
a) To what enzyme class does PGM belong?
b) There are two distinct classes of PGM, one which is dependent on 2,3-bisphosphoglycerate (2,3-BPG), dPGM, and one which is not, iPGM. dPGM uses acid base chemistry and a phosphorylated histidine residue to interconvert 3PG and 2PG. The dPGM reaction proceeds with formation of 2,3-BPG as an intermediate. Propose a mechanism for the dPGM-catalyzed conversion of 3PG to 2PG that is consistent with this information.
c) What is the purpose of 2,3-BPG (i.e., why does dPGM require it)?
The clinical signs of two types of galactosemia—galactokinase deficiency or UDPglucose: galactose 1-phosphate uridylyltransferase deficiency—are markedly different. Although both forms cause gastrointestinal pain after drinking milk, transferase deficiency also causes liver, kidney, spleen, and brain malfunction, as well as mortality. With each kind of enzyme shortage, what products accumulate in the blood and tissues? Estimate the relative toxicity of these goods based on the information provided above.

- The Energy Cost of dTTP Synthesis (Integrates with Chapter 20.) Starting from HCO3, glutamine, aspartate, and ribosc-5-P, how many ATP equivalents are consumed in the synthesis of dTTP in a eukaryotic cell, assuming dihydroorotate oxidation is coupled to oxidative phosphorylation? How does this result compare with the ATP costs of purine nucleotide biosynthesis calculated in problem 2?arrow_forwardAlthough most enzymes are quite specific, they can catalyze side reactions with compounds that are structurally similar to their physiological substrates, but usually at much slower rates. For example, glyceraldehyde- 3-phosphate dehydrogenase (GAPDH), which normally catalyzes the oxidative phosphorylation of glyceraldehyde-3-phosphate, can slowly convert erythrose-4-phosphate, an intermediate in the pentose phosphate pathway, to 1,4-bisphosphoerythronate: Draw a plausible mechanism for this side reaction of GAPDH.arrow_forwardOrotic aciduria is a rare hereditary disorder due to deficient orotate phosphoribosyltransferase and orotidine-5'-decarboxylase activities that are encoded by Uridylate (UMP) synthetase gene in de novo pyrimidine biosynthesis. The characteristic of this disorder is excessive excretion of orotic acid in urine. A mutation of UMP synthetase gene has been identified as R96G (at amino acid position 96, Arginine is changed to Glycine). In orotic aciduria, predict the consequences of deficient UMP synthetase by identifying which downstream enzyme missing its action. One of important molecules to be derived from pyrimidine biosynthesis is deoxycytidine triphosphate (dCTP) for DNA replication and repair. How is dCTP synthesized, include what enzymes and important intermediates in the pathway? 3. As thymidine triphosphate (dTTP) is needed for DNA replication and repair, what enzymes and intermediates are required to get dTTParrow_forward

- Considering the gluconeogenesis that may occur after removal of the amino group from amino acids, will malate be an intermediate in all such metabolic routes between amino acids and glucose? Explain. The phosphoenolpyruvate carboxykinase (PEPCK) gene is more actively transcribed when glucagon is present in blood. Putting the signal cascade responsible for that activation aside for the moment, will the reaction that PEPCK mediates be in zero or first order for that increased transcription to have a maximal affect on gluconeogenesis?
