Chapter 18, Problem 10TYPSS
Summary Introduction

To analyze:

Colonies whose cells might have mutations from the given figure. The sequences must be analyzed for wild type or normal. Predict three types of E.coli colonies found in lactose metabolism mutants. After obtaining a mutant phenotype, the alteration to be performed for the mutant phenotype of these mutant colonies.

Introduction:

The X-gal is a colorless pigment, which can be cleaved by the β-galactosidase enzyme. The pigment is found attached to the galactose. The β-galactosidase enzyme cleaves off the X-gal pigment, which gives a blue-colored appearance.

The following polynucleotide was synthesized and used as a template forpeptide synthesis in a cell-free system from E. coli.                                    …AUAUAUAUAUAUAU… If the same polynucleotide described is used with a mitochondriaderivedcell-free protein-synthesizing system, the product is                                  …Met–Tyr–Met–Tyr–Met–Tyr…What does this say about differences between the mitochondrial and bacterial codes?
Shown below is a schematic diagram of the E. coli leader peptidase (Lep), which has several basic amino acids in a cytoplasmic loop. Propose a mutant of Lep that would be a test of the “inside positive” rule for the orientation of proteins in membranes.
Suppose that the diagram below represents the genomic organization of an enzyme involved in eye pigment production in mice. Within the gene are four exons. Biochemical analysis has revealed that the active site of the enzyme is located in the C terminus of the protein. -The nucleotide length of each exon and intron is shown. -The dinucleotide sequence GT represents the 5’ splice site and the dinucleotide sequence AG represents the 3’ splice site. Both the 5’ and the 3’ splice sites must be present for splicing to occur. Assume that the first and second stop codons are located immediately after the first and second 5’ splice sites, respectively; the third and fourth stop codons are located near the 3’ end of exons 3 and 4, respectively; all these stop codons are in the correct reading frame. a) draw what the processed mRNA will look like. Include the start codon on the mRNA and label the approximate locations of the 5’ UTR and 3’ UTR on the transcript. (You do not need to add the 5’ CAP…
