Chapter 24.9E, Problem 24.23P
Interpretation Introduction

Interpretation:

The structure for vasopressin is to be proposed.

Concept introduction:

Vasopressin is a peptide prohormone that regulates the effective osmotic pressure gradient. It comprises 9 amino acids. Amino acids are carbon containing compounds that possess two functional groups named as amine (NH2) and carboxyl (COOH) and these are prepared by reductive amination of αketoacids by using transaminase enzyme and bromination of carboxylic acid groups followed by amination. Reductive amination comprises transformation of carbonyl group of keto acids to an imine intermediate, which leads to the formation of an amine. Transaminase enzymes are specifically used for speed up the transamination reactions.

An unknown decapeptide was isolated and characterized. Complete hydrolysis of this peptide gave : F(2), A,G,C,K,N,T, W and V. Treatment with carboxypeptidase releases A. Reaction with Edman’s reagent gave PTH-T and a nonapeptide. The nonapeptide was treated with trypsin and gave 2 peptides: (V-C-G-A) and (N-FF-W-K). Give the sequence of amino acid in the decapeptide.
The side chain of acetyl-L-lysine-OMe has a pKa=10.5 and its acid-dissociation reaction  is shown below.  B. At what pH does [A-]=[HA]?
(a) The two most acidic hydrogens of uracil have pKa’s of 9.5 and 14.2, respectively. Match these pKa’s with the hydrogens in the structural formula and provide structures for the most stable resonance contributors of the monoanion and the dianion.(b) The pKa of the conjugate acid of triethylamine is 10.4. Is triethylamine a strong enough base to convert uracil to its monoanion? To its dianion?
