Chapter 12, Problem 24P
Summary Introduction

To analyze:

It is relatively easy to detect gene conversion in four-cell and eight-cell asci of fungi, where ratios such as 3A:1a or 5a:3A indicate that gene conversion has taken place. Why is gene conversion much more challenging to discover in multicellular eukaryotes?

Introduction:

Gene conversion can be defined as a process of so-called directed DNA sequence change that occurs by base-pair mismatch repair within heteroduplex DNA. Gene conversion can switch one allele to another allele already present in a heterozygous genotype. This process is mostly detected in fungi that form ascus such as Neurospora crassa. Ascus is a sack of haploid spores that are the products of meiotic division.

Three haploid fungal mutants that require compound W for growth were isolated. Each mutant contains a recessive allele in a single gene. Three compounds (A, B and C) in the biosynthetic pathway to W are known, but their order in the pathway is unknown. Each compound is tested for its ability to support the growth of each of the three mutants. Phenotypes of all of the three mutants are shown in the following table (“+" indicates growth, "-" indicates no growth). A C W Mutant 1 Mutant 2 Mutant 3 What would be the phenotype of a haploid mutant that contains both mutant alleles in mutant 2 and 3? Phenotype refers to growth or absence of growth on compounds A, B, C and WN. O Like mutant 1 O Like mutant 2 Like mutant 3 O Like wild type
For a haploid fungus, the starting point in the biosynthesis of the amino acid arginine is Compound X, which is always present in and absorbed from the environment. The arginine biosynthetic pathway is: Enzyme A Enzyme B Enzyme Ç Compound X It is know that genes encoding enzymes A and C are on two different chromosomes. Compound Y Compound Z- Arginine A mutant strain of genotype a (lacking only enzyme A) is crossed to a mutant strain of genotype c (lacking only enzyme C) to generate a diploid strain. Sporulation (i.e. meiosis) is subsequently induced in the resulting diploid strain. What proportion of the spores (haploids formed by sporulation) is expected to grow on medium without arginine but supplemented with Compound Y? O 100% 50% 0% 25%
What percentage of the DNA sites in yeast are accessible, assuming that the fraction of sites observed for GAL4 is typical? To how many base pairs of the 12-Mb yeast genome does this percentage correspond?
